i had a build up and running. MSI x99a SLI plus motherboard had windows installed. all was good had mobo about a year with light use. Today i moved the machine to my main desk. i went to turn it on and it poped right off. I looked and there was a fan screw sitting on the motherboard. and i cant get power ever since

Pulled mobo sat it on cardboard out disconnect EVERYTHING but cpu
reset bios ( pulled battery and jumper)
i got a different power supply

when i plug in PSU and flip the power supply switch the motherboard has a small light that turns on blue and then goes off.. using the power button ( on motherboard not case) does nothing. my PSU fan doesnt even start spinning ( yes i have unplugged and reset both sets of power plugs on mobo)

could 1 little screw really kill a mobo?

YES! I had that happen once. I was swapping CPUs, so I removed a mobo (Intel S775) to get at the heatsink, and when I was removing it, one of the standoffs kept turning, or wasn't there, or something.

Well, to make a long story short, I didn't pay enough attention to the fact that something was amiss, and I attempted power-on, and either the screw, or more likely the standoff, was caught under the mobo between the case, and it short it out. Mobo wouldn't boot after that.

It was for a friend, and I had purchased a "backup" mobo for him off of ebay (IIRC), just in case something happened to his. Little did I know that it would be my carelessness killing his original mobo.

Anyways, I got his machine fixed up, but what a PITA. I was careless, and kind of in a hurry, and it bit me.

To be clear, you do not have ram installed and a vid card connected to a monitor while testing right now? Because you need them to boot, and get in the bios, if it does boot.

Mainboards have changed for the better over the years, and a high quality board is now more likely to survive a screw falling on it, or a light screwdriver scratch. If it did manage to fry the board, contact MSI. It has a 3yr warranty, and they may let you RMA despite the, wait for it....screw up.

I put to long of a screw into a Northbridge fan once and fried a board once in the past.

Welcome to oops, oh shit.

I bought another one, that was my fault.

Shorting anything out on a MOBO is not going to have a happy ending.
