Memory interleaving on VP6

In order to see the effects of 2/4-way memory interleaving (set in the bios) on the Abit VP6 motherboard, do I have to have multiple memory banks filled? Is there a difference in performance increase due to interleaving when a single bank is filled vs. when multiple banks are filled?

As a side note, I'm not just sitting here with a working VP6 asking questions I could test on my own 🙂, I'm scoping out different options for a computation cluster my research group is considering.
