We use iMacs at the office I work at, and I'm looking to replace the hard drive in one of them. It's an iMac 10,1 and it has a 500GB something or other in it already. I know in some cases there is a limit to how large of a hard drive can be addressed, would it be alright to put in a 3TB drive in the late 2009 iMac 10,1?

That should work just fine. OS X and HFS+ don't have any (currently feasible) capacity limits, and I can't imagine that the chipset would have any limitations.

Ditto to that. The Core 2 Duo platform (which is what the iMac 10,1 is based on) has no problem with very large hard disks. And since it's a Mac, GPT partitioning (necessary for >2.2TB) is the default anyhow.
