It depends on where they line up. A tailback is called a tailback because they line up the farthest in the backfield. Typically, a fullback lines up before a half or tailback so he can be a lead blocker.

A halfback usually lines up in the wishbone or pro set while a tailback typically lines up in some form of the I

In current NFL no real difference in the designation due to pass nature of the game and lack of multiple back lineups.

In college you still see them being used as the nature of that game allows a running based offense to still be successful.

it varies by team but basically the primary ball carrier is termed the Running Back (RB), Tailback (TB), or Halfback (HB), in order of popularity.

Some specific offensive formations have defined terms for half back, etc.

On a slightly different note, kinda funny how Halfback morphed into Running Back over time, but Fullback never really made the jump to Blocking Back.
