Is a superior signing an email with their first name cue enough...

I've always wondered this. If I'm in contact with a professor, or a boss at work and he / she signs his / her emails "Bob," etc., can I address them by that in a hypothetical reply email?

I'd say that's not enough to address them in person by their first name, but in a correspondence.
 
Go ahead and reply using first name but use "Dear Bob" not "Bob" at first. What I find awkward is when my full name and contact info is in the sig file, sometimes I need to sign with my full name anyway. It seems stiff.
