Usually, with laptops, and phones as well, I let them charge for ~12 hours (maybe 8 depends on how patient/busy I am) and not use it while I'm charging it.
edit: of course, I only do this the first time around.
When you get a new Lithium battery, you need to fully charge it. I have recharged my batteries when necessary without fully discharging and haven't noticed any differences. One obvious situation is that replacement of battery is expected every 1 1/2 to 2 yrs of constant usage.
No matter what, plug it in and charge up the battery for a good long time when it first arrives.
Some people recommend fully charging the battery, then run it off battery for your "normal" applications until the battery goes dead. Doing this once will supposedly calibrate the battery's "time remaining" calculations. Note this has nothing to do with the "memory effect" that oldschool NiCad batteries had.
