According to mapquest, it's:

Total Estimated Time: 4 hours, 36 minutes
Total Distance: 187.84 miles

Is this accurate for travel during mid week?

Depends on what time of the day and is most of it highway driving or...? Even at 60mph that's a little over 3 hours assuming highway the whole trip

I'd say pretty close.

If you try going in on a friday or leaving on sunday night or monday morning it might be a bit longer...

I've only been a couple times. Wasn't too bad though. If there's an accident on the bridge or one of the other bottlenecks, a hurricane coming in, or something (fantasy fest) in town to attract crowds, it could be real bad.

That IS florida driving, right?
Florida driving exacerbated by the fact that the Keys highway is only two lanes so you're screwed if you get stuck behind 15 cars that are backed up behind one slow-mover.

Mid week at this time of year really shouldn't be too bad.
It is a beautiful drive, though, so if you can afford a little more time then it would be worth it, IMO.

Also keep in mind Route 1 to the Keys is the kind of highway that has signs along the side of the road, posting the number of accident deaths the road has claimed this year. We drove to Key Largo Memorial Day weekend, and there was literally a rear-end accident EVERY mile or two. Mostly accidents caused by people slowing down to rubberneck the previous accident: ie. some dude in a pickup slammed into the Mercedes behind me when traffic slowed for an accident ahead. That being said, it CAN be a nice drive. I would give myself the whole 4.5hrs though.
