Here is the lineup:

Frontier Labs Nex IIe

e-digital Treo 10

e-digital mxp-100

MPIO DMG

Here's what I'm going to be using it for:
Running and working out in general
Commuting
Walking around campus

I won't be using the stock headphones so those don't matter. I'd like to get one for as cheap as possible so don't suggest an i-pod. Also, I don't want one that doesn't have the option to upgrade the memory capacity. I also need one that supports variable bit rate and up to 320kbps. I will be using Grado SR-60s and Koss KSC-35s as headphones. Optimally, the unit will have a powerful built-in amp and a high S/N ratio. The Intel Pocket Concert would fit that bill, but it doesn't allow extra memory. The MPIO looks like it has a pretty weak amp, but decent S/N ratio. I'm leaning towards solid-state memory because of running and such, but the Treo can store so much for around the same price.

I've got the MPIO DMG (128 internal, 128 smart card, 256 total) and it kicks ass.

I went with the iPod. It is hard to beat. 20 minute buffer and I have NEVER had a skip when I am running.

Yeah...no ipod.

I think I may have narrowed it down to the DMG and Nex IIe. I was almost set on the IIe, but I did a bit of searching and many people complained about audible distortion at low volume levels. I want the best SQ possible, so that's not acceptable. One reviewer even claimed it had a THD of .5%, which is very high in my opinion. However, some people claim that the newer firmware fixes the distortion problem so who knows.

its not among ur choices, but if found the Rio 500 to be more than satisfactory for my listening needs during running and travelling and what not.

64mb built in, flash-it, add a 128mb card and ur good to go!
