It varies for me what I do after I get started, but at the start I always build a worker first so I can take advantage of any bounus sqaures, next comes a granery to increase growth rate, and then a worker, settler, worker, settler so I can have three cities. Any more than three cities just doesn't seem to work at the start of the game. After that I should have the tech to build spearman so I build barracks and spearman for defense. In my second and third city I build a granery first and another worker second so I can have two workers for each city.
Research order is also very important. Unless you're crowded by a nearby civ you need to research those techs that allow you to gain the benifits of bounus tiles. It also gives you a better idea of where to place your second and third city, ie where is there copper, horses, and even iron.
