From a comfort standpoint the Grado headphones may not be the best. They definitely take a bit of breaking in before they sit comfortably. You will also need to bend the metal band a bit and play with it in order to get it to conform to the shape of your head.
Check out the link below for potential problems with Grado headphones that might cause you to dislike them. It's for the Grado SR325i (I own a pair) but most Grado's are pretty much designed the same way.

The foam ear pads might be uncomfortable for some but you can get ($$) replacements that are softer. One problem is that the weight and design will mean that shaking your head all over the place pretty much means these headphones are going to slip off.
***EDIT
I wear glasses pretty much full time except for outdoor activities when I switch to contacts. These headphones were not really that uncomfortable while wearing glasses. While I have had no time for gaming or anything for quite a while now (baby in the house), I have played games with them for a couple of hours at a time. They are noticeable and i wouldn't say they are real comfortable but I can't say they are annoying.
Again, they do have a break in period and some (like me) may need to adjust the metal band that sits on top of your head. Overall, great headphones though I don't recommend going higher than the Grado SR80 or 125's because for most, going past that is overkill.
