GPO

The GPO is live as soon as you close the GPO editor.

Machines/users will refresh whenever they're ready:
Machines: At every boot (while network connected), at login if using DUN, and it refreshes ~ 12 hours (by default).
DCs refresh more often.
Users: At every logon (when network connected or DUN). Refreshes by default 90minutes. (I think--it's also set by GPO).

If you want to refresh a particular machine, as a local admin on the machine, issue:
secedit /refreshpolicy

Use gpresult 2 minutes later to confirm that the policy updated.
