Why would each of the 2 vectors (one containing the DB Domain and the other the AD) needed for a Yeast 2 Hybrid experiment need to have 2 selectable markers such as Leu2 AND Amp resistance? Seems kind of redundant since one marker should be sufficient for selection?

uh... the amp resistance isn't for the yeast, it's for when you grow it in e.coli and you need to select for the presence of the plasmid. The leu2 is for when you stick it in yeast w/ a leu2 disruption, so now you can select for growth on -leu media.

Oh yea, thats right. Forgot you had to make copies of the plasmid first :)

Thanks
