The answer is a little more complex than yes. You can slipstream SP3 as long as you have at least SP1 installed. If your image was pre SP1, then you will have to slipstream SP1 or SP2 before you can slipstream SP3.
And, another suggestion. If you have IE7 installed, uninstall it before installing SP3. Then reinstall it after SP3 is installed. Otherwise, it will not be uninstallable on your Add Remove Progams menu.
On this page it says that it is recommended, but not required to have SP2 installed first. When SP3 first came available there were some discussions of why this was. I don't think there was an offical answer, but someone did some comparisons and there are certain Windows files that are not contained in SP3, that are in SP2.
