If you dig around online you can find a Win32 executable for the original game. Try it.

I actually downloaded a mod called eDuke32, which is a hi-res pack, and also gave the options to remap the game keys, so you can do WASD, always strafe, and a better mouse look, which made the game much easier to play.

I replayed the game awhile back with eDuke32 as well. I still have my Atomic edition disk. It looked pretty good with the pack, the gameplay of course felt like it could have used a bit more depth but that's just because its from a different era. It was mostly a nostalgia trip for me.
