Firewire 400 v. USB 2.0

Firewire 400, if implemented correctly, is faster with less CPU load than USB 2.0. Many external enclosures don't perform as good as they could - so do your review reading homework before you go buy one.
 
Almost every computer is happy with accepting a USB external drive, whereas, most computers are happy accepting a Firewire drive.

Just for ease of use I would go the USB route.
 
FW400 gets you up to around 37 MB/s with less CPU load than the max. ~28 MB/s you'll see from USB 2.0.
