Haha, I love this game! Very addictive. I've found a few techniques to get all the way to round 100 on medium.
First off, save up and get your first lightning tower by round 9, spending the minimum on other defenses, then selling a missile launcher or two to make up the difference. Always concentrate your most lethal (upgraded) defenses within helicopter range near the center, and route your mazes around large batteries of missile launchers and lightning towers (kill zones), so that enemies have to pass them several times, and helicopters have to fly over them. Make good use of the 'goo' towers to slow enemies down, and again, concentrate them near kill zones so enemies linger there longer. Keep in mind that concentrations of max-upgraded towers (especially missiles) are much more effective than spread out level-one towers.
A fave tactic of mine I call 'pulling the cork'- you can look at it as a bit of a cheat, but I say all is fair in non-paused gameplay. Enemies always take the shortest path through a maze, and you can really use this against them. Build your mazes with an area I call a 'cork'- a spot on the maze usually back near the enemy's side that you can open quickly by selling off a low-value gun tower. Once enemies have advanced past this point, yet not far enough that that they're more than halfway through your maze, they'll turn around and head for the 'cork' area that's now a closer exit. So they'll reverse and go right back through your most lethal kill zones. If they get back to the new opening, simply place another tower before they get through (thus corking the bottle) and they'll have to reverse and go back through the maze yet again. Usually one or two times doing this, and any collection of ground enemies is toast.
As a last defense, you can create another type of 'cork' setup near your base- it can be as simple as a row of towers from top to bottom of the field next to the base, with the opening at whichever end it takes them longest to reach. Hopefully, your main defenses will take out all but a handful of enemies that might slip through- as enemies approach the open end of your base defense, quickly sell the farthest tower at the opposite end, and 'plug' the nearest opening- forcing the enemy once again to travel to the other side of the screen-past your row of defenses (helps to have at least a few goo guns and missile launchers here too- and also within helicopter range). Repeat this before the enemy can travel through the new opening- close it after re-opening the other side. Repeat until the enemy is wiped out.
Anyway, a combination of these kinds of tactics will get you through the medium level.
