I think my favorite would be Eko then Desmond. At points I've liked Locke and other times I didn't care for him.
 
Let's hear your least favorite characters too. That's an easy one for me. Rose and Bernard. Those two make me cringe... so annoying.
 
Hurley and Sawyer. The nicknames Sawyer comes up with always crack me up and Hurley, well, for the laughs too.

Claire and Kate are my favorite babes on the show.

I fucking loathe Jack and Kate both and want them to die in a fire.

edit: I'd probably hate Kate less if she weren't constantly by Jack's side.
 
i like sawyer. his character has changed a lot, and i think the actual actor who plays him has gotten a lot better as the series progressed.

jack used to be cool but now he is annoying.
 
Ben for sure, though I like Juliet quite a bit as well. Miles, Faraday, Captain Frank, and Hurley are also very good.

Least favourite character, of those still around, is Kate.

hurley is great as kind of an off-kilter straight man (he's the only one who points out the absurdity of the events around them).

It's hard for me to decide. I think Josh Holliday? is playing sawyer very well. I love Evangeline, but I'm not a big fan of the kate character, but I think evangeline plays her well. It's just who "kate" is that I don't like to much. She's evillll.

I still think that this whole thing is in Hurley's head. That's why he's the guy who is always pointing out how absurd everything is, why he's the guy with a real world connection to the numbers, why saves the day often, etc..
 
Lapidus has the best lines on the show, and actually acknowledges how ridiculous it all is. But out of that list, I'll go with Locke (as long as it's new MIB Locke and not old whiny/insecure Locke).

As far as hotness: Charlotte > Ilana > Kate > Juliette
