I'm hooking up an amp to my sister's factory stereo. I have to use high level speaker inputs. I'm not sure which wire is + and which is -. Would the one with the stripe be the +?

Black is almost always ground. So yes, black stripe = negative and white stripe = positive.

Are you sure you should be wiring an amplifier if you don't know what's ground and what's live?

actually hooking up the speaker cables backwards won't hurt anything, it will just make the channel out of phase.

edit: well that and your speakers will vibrate backwards, which could potentiall damage them at high volumes.
