Delay in opening software on network server?

Help again guys. We have about 8 computers on a network. The hospital software runs on the server. Each computer has a desktop shortcut for the software. Till yesterday, I could click on it and it would open almost instantaneously. Today on a couple of computers, it takes almost a minute to open. After the software opens it works fast, but any idea why it takes so long to open? All Windows based, and mostly Win 8.1.

If I access other files on the server, there is no delay at all.
