How long does it take for deposited checks to be available for withdraw?

If you need the money ASAP, cash the checks at the bank they were issued from. Take the cash to your bank and deposit it.

Usually checks are available within 24 hrs, though.

Can I still sign them over to someone else for this, or would I need to be there?

Some banks will charge you a fee to cash a check if you are not one of their customers.
