I'm building my new rig next week and I have a few questions for you guys.

Is there anything I need to add or modify on my Antec P182 for optimal cooling? or will everything be fine without changing a thing?

Also, Is one CPU fan enough for a Q6600 with a Thermalright 120 Extreme?

You could add the optional 120 mm fan in front of the middle drive cage. Even if you mount your HDs in the bottom drive cage which gets cooled by the existing fan, you will get some cooling of the vid card with the optional fan at the middle cage.
 
There are two fan mounts in the middle: On the actual Hard Drive cage and behind it (near the front filter) You could add a fan in either location. (120mm)

The stock fans that it comes with are pretty decent. They ARE quite noisy when on HIGH. Around the forums the Yate Loon fans are supposed to be even better.
