What would be the likely problem to this situation?

Car does not run when on D (when starting up). Must start car from 1-2 to get it to run. When the max speed is reached on 2 switch over to D and the car runs fine.
 
Your lockup is locked up. On startup, it's connected straight from the engine to the drive wheels. Pretty much the engine thinks it's in 3rd, and cannot up or down shift.

So the only way to fix it is to take it to a transmission shop.

By the way this is like a 2001 Buick with 250,000 miles.

American car went pretty good with no major problems up until now.

Chances are that the linkage is out of adjustment. Crawl under the car and have someone go through the gears and you see a linkage rotating a bracket on the trans. There should be an adjustment screw in that linkage somewhere.

Take it to a good transmission shop and have them inspect the tranny. A good shop will tell you what the problem is.
