Simple. Double click (left click) on the program's icon. If it gives an error about being a 16 bit application, then it's a 16 bit application. If the program runs ok, then it's 32 bits.
All 32bit x86 CPUs are capable of thunking in 16 bit...including 64 bit CPUs running in 32 bit mode. The problem is that when running in 64 bit mode, 16 bit is not supported. Just because an application runs in 32 bit XP does not mean it is a 32 bit app or that it will run in 64 bit vista.
OP, a sure fire way to tell if you have any 16 bit applications running in windows XP is to open the task manager, and from the processes tab search for a process called NTVDM. It is a 16 bit virtual memory space created for the 16 bit app to run in.
