Can I use 2 8GB DDR3 sticks with 2 4GB sticks?

Yes, it should work, depending on your motherboard, and what CPU you are using.
Some really old motherboards don't like this.

Thanks Elixer! Its a brand new motherboard, ASUS ASUS Z97-K, so I guess it should work then. Any idea whether use of different size modules will slow it down? I know that using one module, instead of two, can, for example.

Different size wont effect speed. Just be sure to put them in the right slots so they are in the same channels with each other. And of course that they are the same voltage. Oh, and if they are different speeds, you will have to run at the lower of the two.
