I have 2 hard drives a 2 DVD players on IDE. I want to add a 3rd HD on my SATA connection. Will it work?
MB is a Asus P4C800E deluxe
HD I'm looking at is a Seagate Barracuda 7200.10 320GB 7200 RPM 16MB Cache SATA 3.0Gb/s

I am not familiar with your motherboard or your power supply.

Having said that, if you have a connection available on your motherboard, all you have to do is hook it up.

See if the Asus P4C800E supports SATA w/o raid. My P4PE does not. So to use a single SATA drive, I had to create an ersatz RAID 0 array of one drive.

If your mobo supports SATA w/o RAID, then it should be rather straight forward. Almost PnP. Enable the SATA ports in BIOS and go from there.
