I have a SATA to USB cable so I can boot from my current C:\ drive while having the new HD accessible as well. What's the best way to make the transfer if I just want everything copied over?

You don't need to clone it in strict sense, you just copy all contents from one drive to another, do not forget hidden files and the system should be able to boot from new drive.
