Question Build a Plex NAS computer or buy a prebuilt NAS ?

What would be better or does not not matter ?

It's better to build it.

However, if you're not experienced building and running/managing server class software, then maybe a pre-built machine would be ideal to start?

Ideally you will want pretty high horse power CPU (single core performance being more important than multi-threading on Plex) and likely explore 10Gig networking. But as the user above mentioned, depends on number of clients, number of streams, content (4k vs not), etc. Also depends on whether this is to serve as a server, or just merely be for Plex purposes and you don't feel like having redundancy, etc. I say this because any system managing a seriously large amount of data may want some redundancy to avoid total rebuilds when one thing goes wrong. Depends on your content and uptime needs.
