How about Neil Patrick harris in the first Harold and Kumar movie.

Also thought it was funny in Austin Powers when the film version of Minnie Me turns out to be Danny Devito.

Unexpected.

From what I've read, Sean Connery's role in The Last Crusade was kept pretty hush-hush. So for a lot of people, the first they knew of him being in the movie was in the theater when he turned around and said "Hi Junior!"

the cgi arnold in terminator salvation was unexpected and pretty lol.
 
Will Ferrell in Wedding Crashers. Vince Vaughan the him are part of "that" comedy group, but very unexpected.
 
Although he is not popular anymore, the lines that Lance Armstrong delivered in Dodgeball were hilarious.
