Best Hotel Rewards Program

Most major chains (mariott, hyatt, hilton) give you the option of earning miles or points, and in some cases both. Personally, I use hyatt because I think their rooms are cleaner and larger than the other chains. Purely personal preference though (hyatt is the most expensive of the three - but work pays for it, so who cares).
If you really want to get free nights, sign up for the credit card deals with your hotel of choice and earn points that way.

Cool, I like that. I'll stay in the cheap ones when I'm paying, and in the fancy ones when work is paying! Thanks!

Can anyone comment on Rewards Network (formerly iDine). Says you get cash back rewards on restaurant dining and hotel stays. Check It

Edit: Darn, a $49 annual fee :|

I've been a Marriott Rewards member for years now.
The best thing to do is sign up for their "specials" when they occur. They might have one that gives you a night free for staying two nights. These seem to be more and more rare as the economy has picked up though. Generally, you are going to have to stay at least eight or nine nights before you have enough points for a free night. You can help yourself by watching which hotels you stay at. You get more points for staying at the Courtyard than you do at their Residence Inn, for example. You can always apply for a Marriott Visa. Every dollar spent is one point. Considering that you probably need 22,000 points for a free evening at the Rhiga Royal in NY then that's a lot of charging!

Choice Members are rewarded with one free night for every two nights booked, can't beat that deal.

Byw, it's Starwood that has the best, most generous hotel program. Starwood as won the Freddie Awards every year. Do your research at Flyertalk, where the travel experts hang out.
