They have a number of different civilizations, each from a different region / time period. Generally, they are unique only in that they get certain advantages over other civs - starting with a specific technology, a single special military unit, or a trait that reflects the national/ethinic character (i.e. increased worker productivity, etc). I don't think there were any specific geographic ties (most games take place in a randomly generated world, so there's no island of Japan)

Game is turn based. You're given a few units and start out sometime at the dawn of civ (4k bc?), and work up until the near future Typically you need to balance scientific research (learning new technology, which allows improvements, units, gvt types, and so on), city building, trading with NPCs, and combat/conquest. There are a number of different ways to win. The game rocks if you can get together some friends.
It's the third in the series. I didn't start until Civ II, but there were usually 1-3 expansion packs per iteration. There were a couple other similar precursor games, and another parallel title by Sid - Alpha Centauri, which dealt with a newborn colony on an alien planet, with futuristic technology and a host of other goodies that rendered it superior to the Civ series. Civ IV is slated for release later this year, IIRC.
