Everything you have recapped is right.
No Errors in logs on individual machine or print server.
- I can even watch the print queue on the print server watch the server recieve the request, spool and then send to the plotter.

I tried a different application on this workstation, just to try and rule out spicer but the same problem exisits.
The plotter supports Tiff, and this setup use to work on this machine up to a few days ago. As well as a similar setup on a co-workers machine still does work.

Strange situation here.
A single user is having trouble plotting Tiff files.
Hardware:
Plotter: HP 1050
Workstation: Custom Build, Windows XP (We would like to replace the machine but the user likes it and wants us to try everything possible before that.)

A quick suggestion because you mentioned backing up files from every PCs every night. Why don't you create everyone a home drive for every employee and have them save important files on it.
My Recommendations:

Also in the future you should develop and maintain a spreadsheet of all used IPs and what machine and location they are being used at. For situtations like this don't happen.
It could be worse, say that PC was off and then you add that server running a mission critical app onto the network...
You will want to hook the serial to serial up, Leaving the ethernet for hosts. You will need a DTE to DCE serial cable, or an expensive Telco simulator.

What software package are you using?
If Outlook 2003 I would click on every indvidual and add them to your contact list, then create a new distribution list and add the members, shouldn't take that long, 1-2min for 2-10 people.
Settings are hard to backup, the first one will save all messages and the folder structure, while the second one will save the rules.
As of settings like viewing panes and what not, I just reset them up after. There may be a way to save them but I don't know.
